DEVELOPMENT AND EVALUATION OF NANOSPONGES-BASED BUCCAL TABLETS FOR DELIVERY OF QUERCETIN USING BOX-BEHNKEN DESIGN

Objective: The goal was to develop a controlled-release formulation of quercetin utilizing the cyclodextrin-based nano-sponges as nanocarrier. Methods: Based on preliminary trials 3-factor, 3-level Box-Behnken design employed. Five types from β-cyclodextrin (NS1-NS5) were purposely designed. Quercetin loaded into by freeze-drying method. prepared characterized and formulated tablets evaluated for weight variation, hardness, friability, disintegration studies, dissolution stability studies. Results: particle sizes quercetin-loaded are in between 36.45 135.27 nm, encapsulation efficiency 42.37 88.44 % drug release% at 6h 53.04 82.64 %. In vitro release studies showed that more than 90 drugs released nano-sponge formulations compared only around 45% free suspension after 24 h. FTIR, DSC XRPD confirmed interaction with nano-sponges. TEM image revealed spherical structure drug-loaded can be retained slowly over time. friability obtained satisfactory results. tablet exhibited maximum 99.75 percent controlled behaviour h, suggested no major significant changes within 6 mo. Conclusion: Cyclodextrin-based superior complexing ability increased solubility poorly soluble made delivery, which reduce dosing frequency.
